Supporting Advanced Organoid and 2D Monolayer Models of the GI Tract

The Advanced Analytics Core offers comprehensive support for culturing epithelial organoids and monolayers—collectively referred to as EpCultures. These in vitro models enable powerful GI research but require specialized techniques for establishment, maintenance, and analysis. Our services provide investigators with access to 3D organoids and unique 2D monolayer cultures, which offer greater flexibility. We also supply high-quality conditioned media and expert consultation to support reproducibility and innovation.


Services Offered

Conditioned Media

Custom formulations optimized for GI epithelial culture:

  • L-WRN Media: Contains L-cell-secreted Wnt3a, R-spondin 3, and Noggin

  • Expansion Media: Maintains intestinal stem cell populations (available on request)

  • Differentiation Media: For driving differentiation by removing growth factors (available upon request)

Human Intestinal Stem Cells (hISCs)

Provision of dissociated hISCs with customizable regional origin:

  • Duodenum, Jejunum, Ileum

  • Ascending, Transverse, or Descending Colon


Multi-Well Cell Expansion

Custom culture services for scaling EpCulture systems.

  • Expansion and cryopreservation of 2D or 3D cultures

  • Support for user-provided or AAC-sourced mouse/human samples

  • Plate formats: Transwells, collagen-coated plates, and others


Organoid and Monolayer Generation

From raw tissue to functional cultures.

  • Isolation of intestinal and colonic crypts

  • Derivation of ISCs from biopsies, resections, or mouse models

  • Establishment of 3D organoids or 2D monolayers
